Develops stabilized alpha-helical peptides, called Helicons, designed to enter cells and bind protein surfaces beyond the reach of conventional small molecules and biologics. The platform combines proprietary peptide chemistry and screening with automated synthesis, physics-based computation, artificial intelligence, and translational and clinical data. It supports direct protein-interaction inhibitors, bifunctional degraders, antibody conjugates, and radioligand payload delivery.
Advances zolucatetide, a direct beta-catenin:TCF inhibitor, in an ongoing trial centered on desmoid tumors and expansion settings across Wnt/beta-catenin-driven disease, while developing prostate-cancer degraders and earlier discovery programs. Research partnerships include Regeneron on certain Helicon programs and ARTBIO on lead-212 radioligand therapies. Operations are based in Cambridge, with clinical activity across U.S. and international sites; clinical and future commercial supply is outsourced.
