Profile

Xilio Therapeutics, Inc.XLOHealth Care

Develops masked immuno-oncology therapies intended to concentrate activity in tumors. The model combines internal pipeline development with collaboration-led programs, including XTX101, XTX301, and XTX501; Gilead holds rights to XTX301 under a license agreement.

Builds masked immuno-oncology therapies intended to activate within the tumor microenvironment, aiming to concentrate anti-tumor activity while limiting peripheral effects. The company develops its own pipeline alongside selected collaboration-led programs.

The active portfolio centers on XTX101, XTX301, and XTX501. It discontinued further monotherapy investment in XTX202 when it reprioritized the portfolio.

Under its XTX301 agreement, Gilead holds an exclusive global license to the candidate and related IL-12 molecules. Xilio remains responsible for development through an initial clinical trial, after which Gilead may take on development and commercialization.

The business depends on clinical development, access to financing, and execution of its Gilead collaboration.
